Methodology

Ammonium Sulfate Precipitation

Used to determine the presence of myoglobinuria.

Specimen Requirements

Minimum of 10 mL of urine from a freshly voided urine in a screw-capped, sterile, plastic urine container. No preservative. Send specimen refrigerated. Maintain sterility and forward promptly. If there is a delay, aliquot the urine into a urine myoglobin transport tube within 1 hour of collection.  These tubes are available by contacting the Core Lab at RMH.

 

Urinary myoglobin is highly unstable unless alkalinized. Even with alkalinization, myoglobin deterioration is variable (approximate averages of 10% at 1 day, 20% at 3 days, and 30% at 7 days).

Urine tests for myoglobin may not be reliable. Presence of hypochlorite or microbial peroxidase or other oxidizing contaminants may cause false-positive reactions. Presence of ascorbic acid (high concentrations) may cause decreased results.
